ردیابی مقایسه‏ای سالمونلا تایفی ‏موریم به روش کشت و Real Time PCR در مرغ‌های منجمد وارداتی

چکیده

گوشت مرغ به عنوان یک غذای ارزان، سالم و دارای پروتئین بالا و چربی کم شناخته شده است. یکی از عوامل اصلی بیماری­های غذازاد، سالمونلا و سروتیپ‌های آن می‏باشد. سالمونلا تیفی موریوم یک معضل بزرگ بهداشتی در جهان می­باشد. در این مطالعه مقایسه‏ای تشخیصی، تعداد 73 نمونه مرغ منجمد وارداتی، به روش کشت و همزمان با پرایمر اختصاصی به روش Real Time PCR برای جداسازی جنس و گونه سالمونلا­ تیفی­موریوم بررسی شد. نتایج نشان داد که از 73 نمونه مرغ منجمد وارداتی درآزمون‏های کشت میکروبی، تعداد 5 نمونه آلوده به جنس سالمونلا تشخیص داده شد (8/6 درصد) و از این تعداد، 2 نمونه سالمونلا تایفی موریوم درآزمون آنتی‏سرم تشخیص داده شد (7/2 درصد). در روش مولکولی با پرایمرهای اختصاصی جنس سالمونلا، 9 نمونه آلوده تشخیص داده شد (3/12 درصد) و از این تعداد، 4 نمونه با پرایمرهای اختصاصی سالمونلا تایفی موریوم مثبت تشخیص داده شد (4/5 درصد). با توجه به نتایج مقایسه­ای بین روش آزمون­ها، در روش Real Time PCR، 8/1 برابر (حدود 2 برابر) نسبت به روش کشت، برتری وجود داشته است. بدین ترتیب در پایش مرغ‏های منجمد وارداتی آلوده به سالمونلا، دقت و سرعت تشخیص Real Time PCR  نسبت به روش سنتی کشت قابل ملاحظه بوده است.

Chicken meat is recognized as a cost-effective, healthy source of high-quality protein with low fat content. One of the primary causes of foodborne illnesses is Salmonella and its various serotypes. Salmonella typhimurium is a major public health issue worldwide. This study evaluates the comparative separation and diagnosis of Salmonella spp. and Salmonella typhimurium in 73 imported frozen chicken samples by culture and simultaneous Real Time PCR with specific primers. The results indicated that, out of the 73 frozen chicken samples analyzed through microbial cultivation for Salmonella detection, 5 samples were identified as Salmonella spp. (6.8%). Among these, 2 samples were specifically diagnosed as Salmonella typhimurium using serological method (2.7%). In molecular method, 9 samples were identified as positive (12.3%), with 4 of these PCR-positive samples confirmed as Salmonella typhimurium (5.4%). Thus, the comparative results between the two testing methods revealed that the Real-Time PCR method was 1.8 times (approximately double) more effective than the conventional cultivation method. In conclusion, the monitoring of frozen imported chickens for Salmonella contamination demonstrated that Real-Time PCR offers significant advantages in terms of accuracy and speed compared to traditional cultivation methods.
