Evaluation of Dot-ELISA method in diagnosis of cattle dicrocoeliasis

  Abstract   Dicrocoeliasis is a parasitic disease caused by digenean tremarode Dicrocoelium dendriticum. The serological diagnosis of this helmenthic disease can be made by different techniques. In the present study the Dot-ELISA (Dot – Enzyme linked immunosorbent assay) was used for serodiagnosis of naturally infected cattle. Adult Dicrocoelium dendriticum obtained from the liver of naturally infected sheep were washed in cold phosphate buffer saline (PBS, pH=7.4) and stored at -20 °c. To detect antibody in sera, somatic and excretory-secretory (ES) antigens of mature trematode were used. Somatic and ES antigens prepared with haemogenization and incubation of mature trematode, respectively. For diagnostic purposes 40 positive and 40 negative cattle sera after confirmed by slaughtered were tested.   Diagnostic sensitivity and specificity values for Dot – ELISA using somatic and ES antigens were: 75.5 %, 77 % and 64.5 %, 87% respectively. Positive and negative predictive values for somatic and ES antigens were 77 %, 75.5 %, and 87 %, 64.5 %. Cross reactivity occurred with hydatid cyst (2 cases) and Fasciola (3 cases). Our findings suggested that using specific antigens for serodiagnosis of dicrocoeliasis in ruminant, particularly sheep in Iran is a primary screening test.
