Isolation and Antifungal Resistance Profiling of Malassezia pachydermatis from Canine Ears Against Azole Class Drugs

Abstract
Malassezia is a group of lipophilic yeasts that are naturally present in the skin and mucosal flora of mammals and are found in about 90% of healthy individuals, but with increased fat, moisture, immune dysfunction, or changes in the microbiome, it can become opportunistic and cause dermatitis and otitis externa, especially in dogs. Isolates of 3 Malassezia pachydermatis samples were collected from clinical specimens and then subjected to laboratory confirmation and diffusion tests in the disk and well to determine drug resistance. The results of the urease test were positive and the catalase test showed bubbles in all 3 samples (100%). The results indicate a stronger inhibitory effect of ketoconazole and itraconazole than other azoles. At a concentration of 25 mg/mL, the mean diameters of the inhibition zones were: ketoconazole 41.9 mm, itraconazole 40.1 mm, miconazole 38.0 mm, clotrimazole 36.5 mm, and fluconazole 27.7 mm, respectively. Itraconazole and ketoconazole (and to some extent miconazole) can be considered as more effective options; however, the final choice of drug should be made according to clinical factors and pharmacokinetic parameters in the host.
