Evaluation of the Effects of Different Concentrations of the Alcoholic Extract of Ferula gummosa on the Regeneration of Damaged Skin Layers in Rats

Abstract
Wound healing is a complex, multi-phase process involving structural regeneration of the epidermal and dermal layers. Herbal compounds with antioxidant and anti-inflammatory properties may enhance tissue repair by improving the wound microenvironment. Ferula gummosa (galbanum), a medicinal plant native to Iran, contains bioactive terpenoid constituents. The present study aimed to histomorphometrically evaluate the effects of different concentrations of the alcoholic extract of F. gummosa on skin layer regeneration in a rat full-thickness wound model.In this experimental study, 90 male Wistar rats were randomly allocated into six groups: control, Eucerin, 2% nitrofurazone, and 5%, 10%, and 20% F. gummosa extract. A 1.5-cm diameter full-thickness circular wound was created on the dorsal region of each rat. Topical treatments were administered daily beginning 24 hours after wound induction. Tissue samples were collected on days 3, 7, and 10. Following fixation in formalin and hematoxylin and eosin (H&E) staining, the mean thickness of the stratum corneum, epidermis, and dermis was measured in a blinded manner using image analysis software. The thickness of all evaluated skin layers increased over time in the treatment groups. The greatest increase was observed in the 20% extract group, while the 5% and 10% groups also demonstrated greater increases compared with the control and nitrofurazone groups. However, the differences among groups were not statistically significant. Overall, the alcoholic extract of F. gummosa, particularly at the 20% concentration, appeared to promote cutaneous tissue regeneration and increase skin layer thickness. Despite the lack of statistical significance, these findings indicate its potential role in enhancing wound healing, warranting further studies with longer follow-up and molecular investigations.
